LC-MS/MS Analysis of Sphingophospholipids Using Metal Free Column
Allentown, PA -- (SBWire) -- 02/25/2019 --Compounds with phosphate groups tend to interact with metallic surfaces in the flow path of an HPLC system. This results in peak tailing, carryover, and insufficient sensitivity.
These effects can be reduced by the use of metal-free HPLC system components, the reduction of metal impurities in the stationary phase material, and utilization of PEEK-lined column hardware. By eliminating/minimizing the exposure of these types of analytes to metal components, peak shape is improved. This can be of particular importance in high sensitivity LC/MS analysis.
Sphingophospholipids are major components forming biological membranes, and they serve as intracellular signaling molecules. It is important to measure the amount of these molecules in biological samples because it greatly influences various metabolic diseases such as obesity, diabetes, and Alzheimer's disease. However, in conventional LC analysis of sphingophospholipids, such as sphingosine-1-phosphate (S1P) and ceramide-1-phosphate (C1P), a phosphate group in those molecules can contribute to significant peak tailing, and thus loss of sensitivity and reproducibility.
An improved method, using YMC-Triart C18 hybrid material in a metal free, PEEK-lined column, was recently reported by YMC. YMC-Triart C18 is an ideal choice of stationary phase, due to its very low metal impurity profile. YMC-Triart PEEK-lined stainless steel columns – which include PEEK column frits – can be used with high pressure PEEK tubing and connectors to create a nominally "metal-free" flow stream between the injector and detector.
The improved method, compared to the use of conventional stainless steel hardware, shows greatly reduced peak tailing, as well as improved peak shape and peak intensity in LC/MS analysis of sphingophospholipids.
